目的探讨舌下腺三叶因子2(TFF2)和三叶因子3(TFF3)在大鼠实验性胃溃疡自愈期间表达的变化。方法雄性SD大鼠分为溃疡组(42只)和正常对照组(6只)。溃疡组在胃前壁近胃窦处将0.01ml冰醋酸注入黏膜下层制作实验性胃溃疡模型,对照组不作任何处理。分别以免疫组织化学S-P法和RT-PCR检测溃疡组和正常组舌下腺TFF2及TFF3基因和多肽表达情况。结果 TFF2及TFF3免疫反应阳性物质主要位于纹状管、闰管和肌上皮细胞,管腔内亦有阳性物质表达。TFF2的积分光密度在溃疡第1天时与正常组相比明显增高,第2天最低,第4、第6天逐渐升高(P<0.01),第10~23天均维持在较高水平(P<0.05);而TFF3的积分光密度在溃疡第1、第2天时接近正常组,第4、第6天逐渐增强并高于正常组(P<0.05),到第10天达高峰(P<0.01),至第23天仍高于正常组(P<0.05)。RT-PCR显示TFF2mRNA和TFF3mRNA的转录情况与相应多肽的表达趋势相似。结论舌下腺TFF2和TFF3基因在大鼠实验性胃溃疡自愈期间表达增高,可能参与胃溃疡愈合过程的调节。
Objective To investigate the changes of TFF2 and TFF3 during sublingual healing in experimental gastric ulcer in rats. Methods Male SD rats were divided into ulcer group (n = 42) and normal control group (n = 6). In the ulcer group, an experimental gastric ulcer model was made by injecting 0.01 ml of glacial acetic acid into the submucosa near the antrum in the anterior gastric wall without any treatment in the control group. The expressions of TFF2 and TFF3 gene and polypeptide in sublingual glands of ulcer group and normal group were detected by immunohistochemical S-P method and RT-PCR respectively. Results The positive TFF2 and TFF3 immunoreactive substances mainly located in the striated duct, the intercalary duct and the myoepithelial cells. The expression of positive substances in the lumen was also observed. The integral optical density of TFF2 was significantly higher than that of the normal group on the first day of ulcer, lowest on the second day, gradually increased on the fourth and sixth days (P <0.01), and remained high at the 10th to 23rd days (P <0.05). The integral optical density of TFF3 was close to the normal group on the 1st and 2nd day of ulcer, and gradually increased on the 4th and 6th day (P <0.05), reaching the peak on the 10th day (P <0.01), still higher than the normal group on the 23rd day (P <0.05). RT-PCR showed that the transcriptional status of TFF2 mRNA and TFF3 mRNA was similar to that of the corresponding polypeptide. Conclusion The expressions of TFF2 and TFF3 genes in sublingual glands during the healing of experimental gastric ulcer in rats may be involved in the regulation of gastric ulcer healing process.
