[目的]探讨FMNL2基因沉默对人大肠癌细胞体内外增殖及侵袭能力的影响。[方法]筛选并建立FMNL2基因沉默的SW80/M5细胞系;荧光定量RT-PCR和Western blot检测干扰效率;CCK8法检测FMNL2对SW80/M5增殖能力的影响;流式细胞仪检测细胞周期;侵袭小室实验检测其对SW80/M5侵袭能力的影响;裸鼠皮下成瘤实验和脾注射法体内转移实验检测FMNL2基因表达沉默对SW80/M5在体内增殖、侵袭能力的影响。[结果]FMNL2基因沉默后显著抑制SW80/M5在体内外增殖、转移及侵袭能力(P<0.05)。[结论]FMNL2与大肠癌多种恶性生物学功能的调控密切相关,可能是潜在的治疗靶点。
[Objective] To investigate the effect of FMNL2 gene silencing on the proliferation and invasion ability of human colorectal cancer cells in vitro and in vivo. [Method] The SW80 / M5 cell line with FMNL2 silencing was screened and established. The interference efficiency was detected by quantitative RT-PCR and Western blot. The effect of FMNL2 on the proliferation of SW80 / M5 was detected by CCK8 assay. The cell cycle was detected by flow cytometry The effects of FMNL2 gene silencing on the proliferation and invasion ability of SW80 / M5 cells in vitro were detected by cell assay in vitro. The effects of FMNL2 gene silencing on SW80 / M5 invasion and invasion were investigated by subcutaneous tumor formation in nude mice and spleen injection in vivo. [Results] The silencing of FMNL2 significantly inhibited the proliferation, metastasis and invasion of SW80 / M5 in vitro and in vivo (P <0.05). [Conclusion] FMNL2 is closely related to the regulation of many malignant biological functions of colorectal cancer and may be a potential therapeutic target.
