目的和方法 :采用全细胞膜片钳技术 ,研究不同浓度 ( 1μmol/L、10 μmol/L、10 0 μmol/L)雌二醇 (estradiol,EST)对豚鼠心室肌细胞L型钙电流 (ICa L)的影响。结果 :①EST明显抑制ICa L,1μmol/L、10 μmol/L、10 0 μmol/LEST分别使峰值ICa L降低 30 .5 % ,5 0 0 % (P <0 .0 1)和 6 8% (P <0 .0 1) ,该作用具有浓度依赖性 ;②EST对ICa L的I V关系曲线未见明显影响 ,其形状和峰值电压保持不变 ;③EST对ICa L的抑制无明显频率依赖关系 ;④EST使通道的激活曲线右移 ,失活曲线左移以及失活恢复时间延长。结论 :EST对豚鼠心室肌细胞L型钙通道具有明显抑制作用
PURPOSE AND METHODS: Whole-cell patch-clamp technique was used to investigate the effects of different concentrations of estradiol (EST) on L-type calcium current in guinea pig ventricular myocytes )Impact. Results: ①EST significantly inhibited ICa L, 1μmol / L, 10μmol / L and 100μmol / LEST, respectively, and decreased the peak ICa L by 30.5%, 500% (P <0.01) and 68% P <0.01), and this effect was concentration-dependent; ②EST had no significant effect on IV curve of ICa L, and its shape and peak voltage remained unchanged; ③EST showed no significant frequency-dependent inhibition of ICa L; ④EST The activation curve of the channel to the right, inactivation curve to the left and deactivation recovery time extended. Conclusion: EST has significant inhibitory effect on L-type calcium channel in guinea pig ventricular myocytes
