目的研究黄芪总提物 (totalextractofastragalosides,TEA)对佐剂性关节炎 (AA)大鼠的治疗作用与其抗氧化作用的关系。方法测量 (AA)大鼠非致炎侧足爪容积和滑膜细胞LPO ;测量兔滑膜成纤维细胞的增殖量 ;间接测量体外超氧阴离子自由基 (O2 ·)和羟自由基 (·OH)产生量。结果TEA对AA大鼠的阶段性治疗 (90mg·kg-1·d -1 ,d15~23 ,ig) ,不仅有抗关节炎作用 ,且可使AA鼠过高的滑膜细胞MDA水平降至正常 ;对低水平氧化应激诱导兔滑膜成纤维细胞的增殖反应 ,TEA(10~40μg·ml-1)有明显抑制作用。TEA(5~80μg·ml-1)对黄嘌呤 (X)_黄嘌呤氧化酶 (XO)体系以及非酶体系产生O2 · 有明显抑制作用 ,高浓度TEA(80μg·ml-1 以上 )对XO的活性还有抑制作用 ;TEA(0.05~160μg·ml-1)对Fenton反应 (Fe2 +_H2O2)产生·OH引起苯甲酸羟化也有明显的浓度依赖性抑制作用。结论TEA对AA鼠的治疗作用可能与其抗氧化作用有关。
Objective To study the relationship between the therapeutic effect of total extracts of astragalus membranaceus (TEA) on adjuvant arthritis (AA) rats and its antioxidation. Methods (A) Rat non-inflammatory side paw volume and synovial cell LPO were measured; proliferation of rabbit synovial fibroblasts was measured; indirect measurement of superoxide anion radical (O2 ·) and hydroxyl radical (OH) in vitro. ) Yield. RESULTS: Staged treatment of AA rats with TEA (90 mg·kg-1·d -1 , d15–23, ig) not only had anti-arthritis effect, but also reduced MDA levels in synovial cells in AA rats. Normal; TEA (10 ~ 40μg · ml-1) significantly inhibited the proliferation of rabbit synovial fibroblasts induced by low-level oxidative stress. TEA (5-80 μg·ml-1) significantly inhibited the production of O2 by the xanthine (X)-xanthine oxidase (XO) system and non-enzymatic systems. High concentrations of TEA (above 80 μg·ml-1) against XO were observed. The activity was also inhibited; TEA (0.05-160 μg·ml-1) produced an hydroxyl radical on the Fenton reaction (Fe2 +_H2O2) and caused hydroxylation of benzoic acid in a concentration-dependent manner. Conclusion The therapeutic effect of TEA on AA mice may be related to its antioxidation.
