用EASYspin Plus Plant RNA试剂盒法、CTAB-LiC1法、异硫氰酸胍(GI)法和TransZol试剂盒法提取了一年生三七绿、紫和绿紫过渡地上茎的总RNA,并借助隶属函数法综合评价了RNA的得率和纯度.结果表明:对于3种地上茎,4种方法所得RNA的得率趋势均为:EASYspin Plus Plant RNA试剂盒法>CTAB-LiCl法>GI法>TransZol试剂盒法,RNA完整性的趋势却为:EASYspin Plus Plant RNA试剂盒法>TransZol试剂盒法>CTAB-LiCl法>GI法,不同方法提取的同种地上茎RNA的OD260/OD280值不相同.提取方法相同时,地上茎颜色制约着所得RNA的得率和OD260/OD280,但与RNA的完整性无关.但是,地上茎相同时,4种方法所得RNA得率间的差异达到极显著水平,但RNA的OD260/OD280值间的差异未达到显著水平;方法相同时,3种地上茎RNA得率间的差异达到显著水平,但RNA的OD26(0/OD280值间的差异未达到显著水平.因此,EASYspin Plus Plant RNA试剂盒法提取一年生三七地上茎RNA的效果最佳.“,”The total RNAs of the green,purple and green-purple transitional aerial stems of one-year-old Panax notoginseng were extracted by using EASYspin Plus Plant RNA kit,CTAB-LiCl,Guanidinium isothiocyanate (GI) and TransZol kit,and subordinate function method was employed to comprehensively evaluate the yields and purity of the RNAs.The results indicated that,for the three aerial stems,the RNA yield tendencies of the four methods were all as EASYspin Plus Plant RNA kit>CTAB-LiCl>GI>TransZol kit,whereas the RNA integrality tendencies were as EASYspin Plus Plant RNA kit>TransZol kit>CTAB-LiCl>GI.The RNA OD260/OD280 values of the same stem extracted by different methods were different.When the extraction methods were the same,the stem colors affected the yields and OD260/OD280 values of the RNAs,but were not related to the RNA integrity.However,when the stems were the same,the differences among the RNA yields of the four methods reached the extremely significant level,but the differences of the RNA OD260/OD280 values were not significant.When the methods were the same,the differences among the RNA yields of the three stems reached the distinct level,whereas the differences among the RNA OD260/OD280 values did not reach significant level.Thus,the EASYspin Plus Plant RNA kit method for extracting total RNAs from the aerial stems of one-year-old P.notoginseng might have the best extraction effectiveness.
