目的:比较罗格列酮钠和马来酸罗格列酮在高果糖诱发胰岛素抵抗大鼠模型上的降糖作用。方法:采用葡萄糖氧化酶法测定高果糖饲料诱发及其后给予胰岛素0.5 U·kg~(-1)对血清葡萄糖含量的影响。用体外放免法测定血清胰岛素水平。结果:60%果糖喂养5周后与正常饲料喂养组比较,高果糖组胰岛素对血清葡萄糖含量影响显著降低。表明已成功建立了高果糖诱导的大鼠胰岛素抵抗模型。罗格列酮钠(3,6,12μmol·kg~(-1))与马来酸罗格列酮(6,12μmol·kg~(-1)),连续给药5周,均能增强胰岛素抵抗大鼠对胰岛素的敏感性,增强胰岛素的降糖作用。并能降低胰岛素抵抗大鼠空腹血清胰岛素水平,作用强度接近。结论:在高果糖诱导的大鼠胰岛素抵抗模型上,罗格列酮钠与马来酸罗格列酮的降糖作用相当。
OBJECTIVE: To compare the hypoglycemic effects of rosiglitazone sodium and rosiglitazone maleate in a rat model of insulin resistance induced by high fructose. Methods: The glucose oxidase method was used to determine the effect of 0.5 U · kg -1 insulin on the serum glucose level induced by high fructose diet. Serum insulin levels were measured by radioimmunoassay in vitro. Results: Compared with normal group, the effect of insulin in high fructose group on serum glucose decreased significantly after 5 weeks of 60% fructose feeding. This indicates that the model of rat insulin resistance induced by high fructose has been established successfully. Sodium rosiglitazone (3,6,12 μmol · kg -1) and rosiglitazone maleate (6,12 μmol · kg -1) were administrated for 5 weeks, all of which increased insulin Resistance to insulin sensitivity in rats and enhance the hypoglycemic effect of insulin. And insulin fasting insulin can reduce fasting serum insulin levels, the intensity of action is close. CONCLUSIONS: Rosiglitazone sodium and rosiglitazone maleate have a comparable hypoglycemic effect on high fructose-induced insulin resistance in rats.
