观察肾上腺髓质素前体N端20肽(PAMP)和肾上腺髓质素(ADM )对血管紧张素II(AngII)诱导的血管平滑肌细胞(VSMC)增殖的影响。用测定培养的大鼠VSMC3H-TdR掺入和蛋白激酶C(PKC)活性的方法。结果发现:10-8mol/L AngII刺激培养的VSMCs 3H-TdR掺入增加2.68倍(P<0.01)、PKC活性增加1.02倍(P<0.01)。而10-9mol/L-10-7mol/L的PAMP或ADM与10-8mol/L AngII同时孵育,则显著抑制AngII的上述作用(P<0.01)。且两者的抑制作用无明显差异。PAMP和ADM均有抑制AngII所致的VSMC增殖作用,可能是通过抑制信号传导系统中的PKC活性来实现的。PAMP和ADM 与AngII相互作用,共同调节VSMC的增殖,从而维持循环系统功能的稳态。
The effects of adrenomedullin precursor N-terminal 20 peptide (PAMP) and adrenomedullin (ADM) on the proliferation of angiotensin II (AngII) -induced vascular smooth muscle cells (VSMCs) were observed. Methods of assaying cultured rat VSMC3H-TdR incorporation and protein kinase C (PKC) activity were used. The results showed that 3H-TdR incorporation increased by 2.68-fold (P <0.01) and PKC activity by 1.02-fold (P <0.01) in VSMCs stimulated with 10-8mol / L AngII. However, incubation of 10-9mol / L-10-7mol / L PAMP or ADM with 10-8mol / L AngII significantly inhibited the above effects of AngII (P <0.01). And the inhibitory effect of the two was no significant difference. Both PAMP and ADM inhibited the proliferation of VSMC induced by AngII, probably by inhibiting PKC activity in signaling system. PAMP and ADM interact with AngII to regulate the proliferation of VSMC and thus maintain the homeostasis of circulatory system function.
