探讨重组水蛭素Ⅲ(rHV3)对半乳糖损伤的人晶状体上皮细胞(hLEC)热应激蛋白(Hsp)表达的影响。采用125×10-3mol/LD-半乳糖建立人晶状体上皮细胞SAR01/04损伤模型,用重组水蛭素Ⅲ干预受损细胞,设置正常组、模型组、和重组水蛭素Ⅲ组,用倒置显微镜观察各组细胞密度和形态,RT-PCR检测人晶状体上皮细胞内3种热应激蛋白mRNA的表达。结果显示,125×10-3mol/LD-半乳糖导致人晶状体上皮细胞显著损伤,3种热应激蛋白,即Hsp70、Hsp27和αB-晶体蛋白的mRNA表达显著上调。重组水蛭素Ⅲ的干预使细胞形态趋于正常,细胞内Hsp70、Hsp27和αB晶体蛋白的mRNA表达显著下调。因此,重组水蛭素Ⅲ能有效缓解高浓度半乳糖导致的热应激蛋白应激耐受,从而保护晶状体上皮细胞。
To investigate the effect of recombinant hirudin Ⅲ on heat shock protein (Hsp) expression in human lens epithelial cells (hLEC) damaged by galactose. Human lens epithelial cells SAR01 / 04 injury model was established by using 125 × 10-3 mol / L-galactose. The damaged cells were treated with recombinant hirudin Ⅲ. The normal group, model group and recombinant hirudin Ⅲ group were set up and observed with inverted microscopy The density and morphology of each group of cells, RT-PCR detection of human lens epithelial cells in three kinds of heat stress protein mRNA expression. The results showed that 125 × 10-3mol / L-galactose resulted in significant damage to human lens epithelial cells. The mRNA expression of three heat-stress proteins, Hsp70, Hsp27 and αB-crystallin, were significantly up-regulated. The recombinant hirudin Ⅲ resulted in the normal cell morphology, and the mRNA expression of Hsp70, Hsp27 and αB in the cells was significantly down-regulated. Therefore, recombinant hirudin Ⅲ can effectively relieve stress tolerance caused by high concentration of galactose, thereby protecting lens epithelial cells.
