作为真核细胞中最重要的细胞器,线粒体不仅在细胞能量代谢中发挥着重要作用,而且是细胞中活性氧(ROS)的主要来源。细胞内ROS在人体生理和病理过程中发挥着重要作用。大量研究表明,ROS与机体衰老、老年痴呆症、心血管疾病、癌症、神经退行性疾病等密切相关。因此,细胞内ROS的研究引起了人们越来越广泛的关注。荧光成像检测由于具有灵敏度高、操作简单、较高的时间和空间分辨率以及对生物体损伤小等优点而成为检测生物活性分子一种强有力的工具。介绍了有机双光子吸收过程,分析了有机双光子线粒体内ROS荧光探针的研究现状,同时对其今后的发展趋势进行了展望。
As the most important organelle in eukaryotic cells, mitochondria not only play an important role in cellular energy metabolism, but also serve as the main source of reactive oxygen species (ROS) in cells. Intracellular ROS plays an important role in human physiology and pathology. A large number of studies have shown that ROS is closely related to body aging, Alzheimer’s disease, cardiovascular diseases, cancer and neurodegenerative diseases. Therefore, the research of intracellular ROS has drawn more and more attention. Fluorescence imaging has become a powerful tool to detect biologically active molecules due to its high sensitivity, simple operation, high temporal and spatial resolution, and small damage to the organism. The process of organic two-photon absorption was introduced, and the current research status of ROS fluorescence probe in two-photon mitochondria was analyzed. Meanwhile, the future development trend of ROS fluorescence probe was prospected.
