为探讨卵泡刺激(FSH)与黄体生成生素(LH)分泌及基因表达与调控用抗人LH-β亚单位单克隆抗体和抗人FSH-β亚单位单克隆抗体进行免疫细胞化学定位的研究。通过连续切片免疫酶技术,免疫细胞化学双标记及电镜免疫金双标记技术,在大鼠垂体前叶进行FSH和LH的免疫细胞化学共定位研究。结果表明:存在着棕(DAB显色示FSH)、蓝(CN显色示LH)及棕蓝混合染色细胞;电镜免疫金双标记发现分别被15nm金粒(示FSH)、10nm(示LH)及被15nm、10nm金粒同时标记的3种细胞(另文报道);光镜连续切片染色亦发现FSH阳性、LH阳性及FSH-LH双阳性3种细胞。因此可以确定垂体前叶存在3种促性腺激素(GTH)细胞,即FSH细胞(只含FSH)、LH细胞(只含LH)和FSH-LH细胞(既含FSH又含LH)。本文对垂体前叶GTH细胞存在着3种激素基因表达形式的可能原因进行了探讨
To investigate the effects of anti-human LH-β subunit monoclonal antibody and anti-human FSH-β subunit monoclonal antibody on follicle stimulating (FSH) and luteinizing hormone (LH) secretion and gene expression and regulation in immunocytochemistry . Immunocytochemistry colocalization of FSH and LH was performed in the anterior pituitary of rat by continuous immunocytochemistry, immunocytochemical double labeling and electron microscopy immunogold labeling. The results showed that there were brown (DAB color display FSH), blue (CN color display LH) and brown-blue mixed staining cells. Electron microscopy immunogold labeling showed that 15 nm gold (FSH), 10 nm (LH) And three kinds of cells labeled with 15nm and 10nm gold particles (reported separately). Three sections of FSH-positive, LH-positive and FSH-LH-positive cells were also found by serial sections of light microscope. Therefore, it was confirmed that there are 3 types of gonadotropin (GTH) cells in the anterior pituitary, namely FSH cells (FSH only), LH cells (LH only) and FSH-LH cells (both FSH and LH).
