目的了解浙江杭州地区乙型肝炎病毒(HBV)基因型分布,初步评价基因芯片技术在HBV基因分型中的应用特点。方法设计多条寡核苷酸探针,用点样仪将探针固定于芯片特定位置上,并与PCR扩增的106例HBV感染者血清HBV基因相应区段杂交,经过显色反应,分析杂交结果,判定基因类型。结果106例HBV感染者中,基因型B者37例,占34.91%,基因型C者69例,占65.09%,未发现其他基因型。结论杭州地区存在HBV基因型B、C型,基因型A、D、E、F是否存在须扩大样本,进一步深入研究。基因芯片技术在同类检测方法中具有灵敏、准确、快速的优势,适用于临床。
Objective To understand the distribution of hepatitis B virus (HBV) genotypes in Hangzhou, Zhejiang Province and to evaluate the application of gene chip technology in HBV genotyping. Methods A number of oligonucleotide probes were designed. The probe was immobilized on specific sites of the chip by dot-matrix and hybridized with the corresponding HBV genome of 106 HBV-infected patients amplified by PCR. After color reaction, Hybridization results, to determine the type of gene. Results Of the 106 cases of HBV infection, 37 cases were genotype B (34.91%), 69 cases were genotype C (65.09%), and no other genotypes were found. Conclusion HBV genotypes B and C, genotypes A, D, E and F exist in Hangzhou area and need to be further expanded. Gene chip technology in the detection of similar methods with sensitive, accurate, fast advantage, suitable for clinical.
