目的 研究去除雄激素对前列腺癌细胞周期调节蛋白表达的影响。方法 在人前列腺癌细胞株LNCaP和PC 3中 ,先加 1× 10 -9mol/L双氢睾酮 (DHT)维持培养 2 4h后分别于去除DTH 0、8和 2 4h行免疫沉淀和WesternBlot分析检测细胞周期素依赖激酶CDK 2、CDK4、CDK6和p2 7kip1的表达。结果 在LNCaP中去除雄激素 8h后CDK2、CDK4、CDK6的表达开始降低 ;而p2 7kip1的表达开始升高 ,到 2 4h时CDK 2、CDK 4、CDK6的表达达到最低 ,而p2 7kip1的表达达到最高。在PC 3细胞中未见CDK2、CDK 4、CDK 6和 p2 7kip1的表达改变。结论 CDK2、CDK4和CDK6的表达降低及 p2 7kip1的表达升高是手术或药物去势治疗雄激素依赖性前列腺癌可能的机制之一
Objective To study the effect of androgen deprivation on the expression of cyclin-regulated protein in prostate cancer cells. Methods The human prostate cancer cell lines LNCaP and PC 3 were cultured and cultured for 24 hours by adding 1 × 10 -9 mol / L dihydrotestosterone (DHT). Immunoprecipitation and Western Blot analysis were performed on DTH at 0, 8 and 24 h, respectively Cyclin-dependent kinase CDK 2, CDK4, CDK6 and p2 7kip1 expression. Results The expression of CDK2, CDK4 and CDK6 began to decrease after androgen was abolished in LNCaP for 8h, while the expression of p2 7kip1 began to increase. The expression of CDK2, CDK4 and CDK6 reached the lowest at 24 h, while the expression of p2 7kip1 reached highest. No CDK2 was seen in PC3 cells, and the expression of CDK4, CDK6 and p2 7kipl was altered. Conclusions The decreased expression of CDK2, CDK4 and CDK6 and the increased expression of p2 7kip1 may be one of the possible mechanisms of surgical or drug castration in the treatment of androgen-dependent prostate cancer
