目的探讨氢氧化铝吸附炭疽杆菌重组保护性抗原(Recombinant protective antigen,rPA)的影响因素。方法取含不同PO43-浓度的PBS和添加EDTA的PBS,分别稀释rPA后,加入氢氧化铝,放置2~8℃及(23±2)℃作用不同时间,检测A280和A260值,计算未吸附的rPA蛋白量,并比较不同溶液中氢氧化铝对rPA的吸附效果。结果氢氧化铝对rPA的吸附率随PO43-浓度的增加而降低;PBS中添加EDTA后,氢氧化铝对rPA的吸附效果明显下降;2~8℃较(23±2)℃的吸附率略有增加,但不同时间的吸附量无明显差异;在H2O中氢氧化铝对rPA的吸附效率比在PBS中高。结论稀释缓冲液和离子强度、EDTA均可影响氢氧化铝与rPA的吸附效果,但吸附温度和时间对吸附的影响不大。
Objective To investigate the factors that affect the adsorption of recombinant protective antigen (rPA) by aluminum hydroxide on Bacillus anthracis. Methods PBS containing different concentrations of PO43 and PBS with EDTA were added. After diluted with rPA, aluminum hydroxide was added to the solution and incubated for 2 ~ 8 ℃ and (23 ± 2) ℃ for different time. Of rPA protein, and compare the adsorption of rPA by aluminum hydroxide in different solutions. The results showed that the adsorption rate of rPA by aluminum hydroxide decreased with the increase of PO43- concentration. The adsorption of rPA by aluminum hydroxide decreased obviously after adding EDTA in PBS. The adsorption rate of rPA at 2 ~ 8 ℃ was slightly lower than that of (23 ± 2) ℃ But there was no significant difference in the amount of adsorption at different times. The adsorption efficiency of aluminum hydroxide to rPA in H2O was higher than that in PBS. Conclusion The dilution buffer, ionic strength and EDTA all affect the adsorption of aluminum hydroxide and rPA, but the adsorption temperature and time have little effect on the adsorption.
