研究红曲代谢产物Monascin对人恶性黑色素瘤细胞A375的诱导分化作用。用MTT法评价Monascin对A375增殖的抑制作用;PI流式单染检测细胞周期分布;透射电镜观察细胞超微结构变化;以黑色素含量、酪氨酸酶活力和体外迁移能力为指标检验Monascin对A375诱导分化作用。Monascin对A375具有一定的增殖抑制作用,并能将其阻滞在G0/G1期;透射电镜观察到A375核质比变小且细胞器增多;黑色素含量增多,相对酪氨酸酶活力提高,体外迁移能力下降。试验结果表明,Monascin对A375具有较显著的诱导分化作用。
To study Monatin inducing differentiation of human malignant melanoma cell line A375. The inhibitory effect of Monascin on the proliferation of A375 cells was evaluated by MTT assay. The cell cycle distribution was detected by PI flow cytometry. The ultrastructural changes of cells were observed by transmission electron microscopy. The effect of Monascin on A375 was assayed by Melanin, tyrosinase activity and migration in vitro Induction of differentiation. Monascin could inhibit the growth of A375 to a certain degree and block it in the G0 / G1 phase. Transmission electron microscopy showed that the ratio of A375 nucleus to cytoplasm became smaller and the number of organelles increased. The experimental results show that Monascin A375 has a more significant induction of differentiation.
