方法:通过免疫组化方法,检测30例正常胃窦黏膜,30例不典型增生胃黏膜和74例胃腺癌组织中VIP蛋白的表达,摘要目的:研究胃腺癌中是否存在分泌血管活性肠肽(VIP)的肿瘤细胞,探讨VIP在胃腺癌发生中的作用。以及中分化胃腺癌细胞系SGC-7901,低分化胃腺癌细胞系BGC-823及未分化胃腺癌细胞系HGC-27中VIP蛋白的表达。结果:36/74例(48.6%)胃腺癌组织中发现VIP阳性细胞,其中高分化7例,中分化15例,低分化14例。阳性(10.0%)正常胃窦黏膜存在VIP阳性细胞。胃腺癌细胞株VIP蛋白呈高水平表达,其中SGC-7901阳性率为71.5%,细胞形态不规则,多数为细胞浆着色,少数细胞膜着色,核大而不规则。4/30例(13.3%)不典型增生及3/30例BGC-823阳性率为82.3%,HGC-27阳性率为92.8%,VIP免疫反应阳性颗粒主要出现在胞浆。结论:胃腺癌组织的肿瘤细胞有较高VIP蛋白表达;胃腺癌细胞株分泌VIP蛋白,且阳性率有随其分化程度降低而升高的趋势。推测VIP蛋白可能与胃腺癌的发生有密切的关系。
Methods: The expressions of VIP protein in 30 cases of normal gastric mucosa, 30 cases of atypical hyperplasia gastric mucosa and 74 cases of gastric adenocarcinoma were detected by immunohistochemistry. Abstract Objective: To investigate whether VIP protein is secreted by gastric adenocarcinoma VIP) tumor cells, to explore the role of VIP in gastric adenocarcinoma. And the expression of VIP in moderately differentiated gastric adenocarcinoma cell line SGC-7901, poorly differentiated gastric adenocarcinoma cell line BGC-823 and undifferentiated gastric adenocarcinoma cell line HGC-27. Results: VIP positive cells were found in 36/74 cases (48.6%) of gastric adenocarcinoma tissues, among which 7 cases were well differentiated, 15 cases were moderately differentiated and 14 cases were poorly differentiated. Positive (10.0%) normal gastric mucosa presence of VIP positive cells. The expression of VIP protein in gastric adenocarcinoma cell line was high, in which the positive rate of SGC-7901 was 71.5%. The cell morphology was irregular, most of which were stained with cytoplasm. A few of the cell membrane were stained and the nucleus was large and irregular. The positive rate of HGC-27 in 4/30 (13.3%) cases of atypical hyperplasia and 3/30 cases of BGC-823 was 82.3%, and the positive rate of HGC-27 was 92.8%. VIP immunoreactive granules mainly appeared in cytoplasm. Conclusions: The tumor cells in gastric adenocarcinoma have higher expression of VIP protein. The gastric adenocarcinoma cell line secretes VIP protein, and the positive rate tends to increase with the degree of differentiation. Speculated that VIP protein may have a close relationship with the occurrence of gastric adenocarcinoma.
