整合素(Integrin)是一大族调节细胞基质以及细胞-细胞相互作用的细胞粘附分子.肿瘤转移是一复杂多步骤过程,其中涉及细胞粘附这一重要先决条件.细胞粘附降低和侵袭性恶性表型可能与整合素受体负调节(下调)有关.整合素是一糖蛋白异二聚体,含有α和β亚属、在恶性肿瘤,丧失整合素表达易导致肿瘤转移,为此作者检查正常、良性和恶性乳腺肿瘤组织中β_1、β_3、β_4和β_5整合素的表达(均为腺上皮中主要的整合素)以观察其与淋巴结转移的关系.取12例良性和61例恶性(50例导管癌,11例小叶癌)乳腺肿瘤的冰冻切片标本,用针对β_1、β_3、β_4和β_5整合素的单克隆抗体作抗生物蛋白一生物素复合法(ABC法)染色,由两位评估者分别读片,取得一致的评分结果.在12例良性乳腺病变中,平均年龄49.4岁,组织学检查半数为正常,乳腺纤维腺瘤和纤维囊性增生各3例.在61例恶性乳腺肿瘤中,平均年龄56.1岁;肿瘤大小平均2.4cm;组织学病期分级,1级14人,2级25人,3级22人;腋淋巴结阳性26人(43%),受侵淋巴结平均数4.5个
Integrins are a large group of cell adhesion molecules that regulate cell matrices and cell-cell interactions. Tumor metastasis is a complex multi-step process involving an important prerequisite for cell adhesion. Reduced cell adhesion and invasiveness The malignant phenotype may be associated with negative regulation (downregulation) of integrin receptors. Integrins are heterodimers of glycoproteins containing subfamilies of α and β. In malignant tumors, loss of integrin expression can lead to tumor metastasis. The expression of β_1, β_3, β_4, and β_5 integrins in normal, benign, and malignant breast tumors (all of which are major integrins in glandular epithelium) was examined to observe their relationship with lymph node metastasis. Twelve benign and 61 malignant cases were taken. Frozen section specimens of breast cancer of 50 ductal carcinomas and 11 lobular carcinomas were stained with anti-biotin complex (ABC) staining with monoclonal antibodies against β_1, β_3, β_4, and β_5 integrins. The evaluators read the films separately and obtained consistent scoring results. Among the 12 cases of benign breast lesions, the average age was 49.4 years, half of histological examinations were normal, 3 cases of breast fibroadenoma and fibrocystic hyperplasia, and 61 cases of malignant mammary glands. In the tumor, the average age was 56.1 years; tumor size was average 2.4cm; histological stage was graded, grade 14 was 14; grade 2 was 25; grade 3 was 22; axillary lymph node positive was 26 (43%), mean lymph node invasion 4.5
