目的:研究壁虎醇提物(Gecko alcohol extract,GAE)对人食管癌EC-109细胞的增殖抑制作用及其机制。方法:MTT法检测GAE对细胞增殖抑制作用;Hoechest 33342荧光染色观察细胞形态学的改变;流式细胞术检测凋亡率;SP免疫组化法检测Fas和Caspase-3蛋白的表达。结果:GAE具有抑制食管癌EC-109细胞增殖的作用;3.0mg/mL、4.0 mg/mL GAE作用细胞48 h后凋亡率分别为20.60%和37.93%;与正常对照组相比GAE组Fas蛋白和Caspase-3蛋白的表达显著增加(P<0.05)。结论:GAE能够抑制EC-109细胞增殖并诱导其凋亡,其作用机制可能与增加Fas蛋白和Caspase3蛋白的表达有关。
Objective: To study the inhibitory effect of gecko alcohol extract (GAE) on the proliferation of human esophageal carcinoma EC-109 cells and its mechanism. Methods: The inhibitory effect of GAE on cell proliferation was detected by MTT assay. The morphological changes were observed by Hoechest 33342 staining. The apoptosis rate was detected by flow cytometry. The expression of Fas and Caspase-3 protein was detected by SP immunohistochemistry. Results: GAE could inhibit the proliferation of esophageal EC-109 cells. The apoptotic rates of GAE treated with 3.0 mg / mL and 4.0 mg / mL GAE for 48 h were 20.60% and 37.93%, respectively. Compared with normal control group, The protein and Caspase-3 protein expression increased significantly (P <0.05). CONCLUSION: GAE can inhibit the proliferation and induce the apoptosis of EC-109 cells. Its mechanism may be related to the increase of the expressions of Fas and Caspase3 proteins.
