目的采用间断小剂量油酸注射方法,构建符合肺损伤标准、血流动力学稳定的乳猪急性肺损伤动物模型。方法中华小型猪6只,经右心耳插管注射油酸-乙醇溶液0.1mL/kg,0.1mL/次,间隔90s直至注射完毕。注射过程中监测循环、呼吸参数;实验结束取右下肺组织标本行组织病理检查。结果油酸注射前、后心率、平均动脉压、平均肺动脉压、心输出量、动脉血氧分压、动脉血二氧化碳分压、pH值、氧合指数差异均有统计学意义(P<0.05)。组织病理显示双肺呈弥漫性改变。结论小剂量间断注射油酸可构建血流动力学稳定并符合肺损伤诊断标准的急性肺损伤动物模型。
Objective To establish a animal model of acute pig-lung injury in piglets that meets the criteria of lung injury and hemodynamics by intermittent small-dose oleic acid injection. Methods Six Chinese miniature pigs were injected with 0.1 mL / kg oleic acid-ethanol solution through the right atrial appendage at intervals of 90 seconds until the injection was completed. During the injection, the circulation and respiratory parameters were monitored. At the end of the experiment, the right lower lung tissue samples were taken for histopathological examination. Results Before and after oleic acid injection, heart rate, mean arterial pressure, mean pulmonary arterial pressure, cardiac output, arterial partial pressure of oxygen, partial pressure of arterial carbon dioxide, pH and oxygenation index were all significantly different (P <0.05) . Histopathology showed diffuse changes in the lungs. Conclusion Small doses of intermittent injection of oleic acid can establish an acute lung injury animal model with stable hemodynamics and diagnostic criteria of lung injury.
