目的观察芎归六君子汤对慢性阻塞性肺疾病(COPD)气虚血瘀痰阻证模型大鼠气道Muc5ac水平的影响。方法60只大鼠按随机数字表随机分为空白组、芎归六君子汤自身对照组、模型组、芎归六君子汤干预组(低、中、高剂量),每组10只。采用改良的香烟烟熏和脂多糖气管注入法联合制备模型。采用芎归六君子汤干预15天后取材,观察肺泡灌洗液(BALF)中Muc5ac水平、气道上皮Muc5ac蛋白表达及肺组织Muc5ac mRNA表达。结果与空白组比较,模型组大鼠BALF中的Muc5ac水平及气道上皮Muc5ac蛋白的表达增加,肺组织Muc5ac mRNA表达上调(P<0.05)。芎归六君子汤能降低COPD气虚血瘀痰阻证模型大鼠BALF中的Muc5ac水平、气道上皮Muc5ac蛋白表达,下调肺组织Muc5ac mRNA表达,且以高剂量组较为明显(P<0.05)。结论芎归六君子汤在一定程度上降低COPD气虚血瘀痰阻证模型大鼠气道Muc5ac水平,减少气道黏液高分泌。
Objective To observe the effect of Xionggui Liujunzi Decoction on airway Muc5ac in chronic obstructive pulmonary disease (COPD) rat model of Qi deficiency, blood stasis and phlegm blocking. Methods Sixty rats were randomly divided into blank group, XGG Liu self-control group, model group and XGHR intervention group (low, medium and high dose) according to random number table. A modified cigarette smoke and lipopolysaccharide tracheal injection method was used to prepare the model. Fifteen days after Xionggui Liujunzi decoction was taken, Muc5ac level, Muc5ac protein expression in airway epithelium and Muc5ac mRNA expression in lung tissue were observed in bronchoalveolar lavage fluid (BALF). Results Compared with the blank group, Muc5ac level in BALF and Muc5ac protein expression in airway epithelium increased in model group, and Muc5ac mRNA expression in lung tissue was increased (P <0.05). Xionggui Liujunzi Decoction can reduce the level of Muc5ac in BALF, expression of Muc5ac in airway epithelium and Muc5ac mRNA expression in lung tissue of COPD model rats with deficiency of Qi and blood stasis and phlegm blocking syndrome, especially in high dose group (P <0.05). Conclusion Xiongguiliujunzi Decoction can reduce the level of Muc5ac in the airway of COPD model rats with Qi deficiency, blood stasis and phlegm blocking to a certain extent and reduce the airway mucus hypersecretion.
