目的:观察赤雹果总有机酸(TOATDF)的解热作用,并探讨其机制。方法:SD大鼠ip内毒素(LPS)100μg·kg-1制备大鼠发热模型。将模型大鼠随机分为TOATDF 300,150,75 mg·kg-1剂量组,阿司匹林150 mg·kg-1对照组及模型组,另取10只大鼠作为正常对照。药后每1 h测量体温1次,连续6次。采用酶联免疫测定法(ELISA)法测定血清中前列腺素E2(PGE2)、肿瘤坏死因子-α(TNF-α)和白介素(IL)-1β及脑脊液中PGE2表达水平。结果:TOATDF 300,150 mg·kg-1剂量组大鼠各时间点的平均体温均低于模型组(P<0.05或P<0.01);75 mg·kg-1组大鼠药后2~5 h体温明显低于模型组(P<0.05);阿司匹林组药后1~5 h体温明显低于模型组。各给药组血清中TNF-α,PGE2,IL-β及脑脊液中PGE2的表达水平均显著低于模型模型组(P<0.05或P<0.01),但高于正常对照组(P<0.05)。结论:TOATRF有明显的解热作用,并有一定的剂量-效应关系。其解热作用与抑制致热原诱发的PGE2,TNF-α和IL-β等致热因子的产生或释放有关。
Objective: To observe the antipyretic effect of TOATDF and its mechanism. Methods: Rat model of fever was prepared by intraperitoneal injection of 100μg · kg-1 of LPS. The model rats were randomly divided into TOATDF 300,150,75 mg · kg-1 dose group, aspirin 150 mg · kg-1 control group and model group, another 10 rats as normal control. 1 h after treatment, the body temperature measured 1, 6 consecutive times. Serum levels of prostaglandin E2 (PGE2), tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) and cerebrospinal fluid levels of PGE2 were measured by enzyme-linked immunosorbent assay (ELISA) Results: The body temperature of rats in TOATDF 300 and 150 mg · kg-1 groups were lower than those in model group at each time point (P <0.05 or P <0.01) Significantly lower than the model group (P <0.05). The body temperature of aspirin group was significantly lower than that of the model group at 1 to 5 h after treatment. The levels of TNF-α, PGE2, IL-β in serum and the levels of PGE2 in cerebrospinal fluid in each administration group were significantly lower than those in model group (P <0.05 or P <0.01), but higher than those in normal control group . Conclusion: TOATRF has obvious antipyretic effect, and has some dose-effect relationship. Its antipyretic effect is related to the inhibition of pyrogen induced PGE2, TNF-α and IL-β and other pyrogen production or release.
