胸腺嘧啶核苷酸合酶(Thymidylate synthase)的催化作用通过有序机制(Ordered sequential mechanism)进行。在不同浓度的蝶酰单谷氨酸衍生物(CH_2-H_4PteGlu_1)存在时,测定与(5-~3H)dUMP的消耗相关的V/K效应,並通过一偶联体系,测定与(5-~3H)dUMP和(2-~(14)C)dUMP混合物的消耗相关的V/K效应.结果表明,~3H对胚胎猪肝胸腺嘧啶核苷酸合酶的动力学效应,与以相同底物所进行的产物抑制作用和死端抑制作用而确立的底物加入和产物释出的次序相符。
The catalytic effect of thymidylate synthase is achieved by the Ordered sequential mechanism. The V / K effect associated with consumption of (5- ~ 3H) dUMP was determined in the presence of different concentrations of the piacetylmonoglutamate derivative (CH_2-H_4PteGlu_1) and determined by coupling reaction with (5- ~ 3H) dUMP and (2- ~ (14) C) dUMP mixture.The results showed that the kinetic effect of ~ 3H on thymidine synthase Substance inhibition and dead-end inhibition were established by substrate addition and product release in the same order.
