目的:观察芍药内酯苷(AF)、芍药苷(PF)对慢性束缚应激(CRS)配合孤养肝郁模型大鼠行为及下丘脑-垂体-肾上腺(HPA)轴的影响,探讨白芍柔肝解郁作用机制及物质基础。方法:将SD雄性大鼠,根据蔗糖水消耗量及体质量随机分组,每组12只。除空白组外,其余均采用CRS配合孤养复制肝郁模型。观察体质量,行为学指标,并用放射免疫法(RIA)检测大鼠下丘脑促肾上腺皮质激素释放素(CRH)、血浆促肾上腺皮质激素(ACTH)、皮质酮(CORT)的含量。结果:造模第14天,与模型组比较,AF 30mg/kg组、PF 30mg/kg组大鼠体质量明显增加(P<0.05,P<0.01)。造模第21天,与模型组比较,AF 30mg/kg组、PF 30mg/kg组大鼠体质量,旷场实验中穿行次数明显增加(P<0.05,P<0.01)而滞留时间显著减少(P<0.01),高架十字迷宫实验OE和OT显著增加(P<0.05,P<0.01),而CT显著减少(P<0.01)。AF 30mg/kg组、PF 30mg/kg组大鼠下丘脑CRH、血浆ACTH、CORT的含量均显著降低(P<0.05)。结论:AF和PF作为白芍的主要有效成分,不仅可在一定程度上改善了CRS肝郁模型大鼠的行为表现,而且使相关应激激素的含量变化呈现不同的趋势,提示二者均为白芍柔肝解郁作用的有效成分,其作用机制与抑制CRS肝郁模型大鼠HPA轴的激活有关。
OBJECTIVE: To observe the effects of paeoniflorin (AF) and paeoniflorin (PF) on behavior and hypothalamic-pituitary-adrenal (HPA) axis in chronic restraint stress (CRS) Rougan Jieyu mechanism of action and material basis. The contents of CRH, ACTH and CORT in hypothalamus in AF 30mg / kg and PF 30mg / kg groups were significantly decreased (P <0.05).
