为获得无核葡萄品种‘Kismis’无涩味后代,以该品种杂交所得幼胚为试材,研究其胚挽救技术。结果表明:通过试验确定采样最佳时期为授粉后41~48 d;通过培养基及不同植物生长调节剂浓度及组合试验筛选出适宜其胚珠发育的培养基为:N69+1.0 mg/L IBA+0.1 mg/L 6-BA+0.5mg/L GA3,发育率最高为55.4%,3种植物生长调节剂因子对胚发育的影响为:GA3>6-BA>IBA;适宜胚萌发培养基为:1/2MS+1.0 mg/L IAA+0.5 mg/L GA3+0.2 mg/L IBA,胚萌发率最高为26.2%,2种植物生长调节剂对胚萌发的影响为:GA3>IAA;胚培苗移栽成活率在70%以上。
In order to obtain the astringent offspring of ’Kismis’, a seedless grape variety, the embryo rescue technique was studied with the immature embryos obtained from the cross of this variety. The results showed that the optimum sampling period was 41-48 days after pollination. The medium suitable for ovule development was screened out by N69 + 1.0 mg / L IBA + 0.1 mg / L 6-BA + 0.5 mg / L GA3, the highest rate of development was 55.4%. The effect of three plant growth regulators on embryo development was GA3> 6-BA> IBA. The highest germination rate was 26.2% with 1 / 2MS + 1.0 mg / L IAA + 0.5 mg / L GA3 + 0.2 mg / L IBA. The effect of two plant growth regulators on embryo germination was GA3> IAA. Transplanting survival rate of 70% or more.
