克隆、分离在致动脉粥样硬化因素作用下血管内皮细胞基因的差异表达对了解动脉粥样硬化发生的分子机制至关重要 ,用改进的差异显示 -多聚酶链技术分离、克隆氧化型低密度脂蛋白刺激培养的内皮细胞有差异表达的基因 ,用Northern印迹法证实差异表达的基因。结果发现氧化型低密度脂蛋白诱导培养的人脐静脉内皮细胞中出现一个上调节的cDNA片段 ,长 2 83bp ,其序列与人胸腺素 β4基因有 10 0 %的同源性 ,具有完整的开放阅读框架。Northern印迹分析证实氧化型低密度脂蛋白诱导人脐静脉内皮细胞胸腺素β4升调了 3倍。从而提示氧化型低密度脂蛋白刺激血管内皮细胞胸腺素 β4的高表达 ,进而调节血管内皮细胞的增殖、分化。改进的差异显示 -多聚酶链技术具有便捷、特异性高、重复性好的特点
Cloning and Isolation of Genes Differentially Expressed in Vascular Endothelial Cells Under Atherogenic Factors is Important for Understanding the Molecular Mechanism of Atherosclerosis. By means of improved differential display - polymerase chain reaction, cloning of oxidized low-density lipids Proteins stimulate differentially expressed genes in cultured endothelial cells and differentially expressed genes were confirmed by Northern blotting. The results showed that an up-regulated cDNA fragment was found in oxidized low-density lipoprotein-induced human umbilical vein endothelial cells, which was 2 83 bp in length with a sequence homology of 10 0% with the human thymosin β4 gene and had a complete open Reading frame. Northern blot analysis confirmed that oxidized low-density lipoprotein induced a 3-fold up-regulation of thymosin β4 in human umbilical vein endothelial cells. It suggested that oxidized low density lipoprotein (LDL) stimulated the high expression of thymosin β4 in vascular endothelial cells, which in turn regulated the proliferation and differentiation of vascular endothelial cells.
