目的研究木脂素对胃癌细胞抗增殖过程中c-myc基因及蛋白的作用。方法实验所用细胞为人胃癌MGC-803细胞,实验分为实验组和对照组,MTT法检测细胞生存率,免疫细胞化学、RT-PCR及Western检测c-myc基因及蛋白的表达。结果实验组人胃癌MGC-803细胞生存率时间和浓度依赖性降低(P<0.01)。在24,48和72 h实验组胃癌细胞c-myc蛋白的表达均呈阴性,而对照组均呈强阳性(P<0.01)。RT-PCR及Western blot结果,c-myc基因及蛋白的表达在对照组胃癌细胞中随时间的延长逐渐增强,而实验组胃癌细胞中逐渐降低(P<0.05)。结论木脂素对胃癌细胞具有明显的抗增殖作用,其机制之一是通过下调c-myc基因和蛋白的表达,从而抑制肿瘤细胞增殖和调控细胞周期。
Objective To study the effect of lignans on c-myc gene and protein in anti-proliferation of gastric cancer cells. Methods The cells used in this study were human gastric cancer MGC-803 cells. The experiment was divided into experimental group and control group. The cell survival rate, immunocytochemistry, RT-PCR and Western blot were used to detect the expression of c-myc gene and protein. Results The survival rate of human gastric cancer cell line MGC-803 decreased in a time and concentration-dependent manner (P <0.01). The expression of c-myc in gastric cancer cells was negative at 24, 48 and 72 h, while the control group was strongly positive (P <0.01). The results of RT-PCR and Western blot showed that the expression of c-myc gene and protein in gastric cancer cells in control group increased gradually with time, but gradually decreased in gastric cancer cells in experimental group (P <0.05). Conclusion Lignans have a significant antiproliferative effect on gastric cancer cells. One of the mechanisms is that lignans can inhibit tumor cell proliferation and regulate cell cycle by down-regulating c-myc gene and protein expression.
