目的 研究微丝功能对正常皮肤及增生性疤痕成纤维细胞胶原酶及金属蛋白酶组织抑制因子 1 mRNA表达水平的影响。方法 采用细胞培养, Northern blot分析等方法,以α 32P dCPT标记的胶原酶及 TIMP 1 cDNA为探针,检测正常皮肤及增生性瘢痕成纤维细胞胶原酶及金属蛋白酶组织抑制因子 1mRNA表达水平的变化。结果 用细胞松弛素 B破坏微丝后,正常皮肤及增生性瘢痕成纤维性细胞的胶原酶及 TIMP 1 mRNA含量明显升高。 结论 微丝骨架可能在基因转录水平参与对成纤维细胞合成细胞外基质的调控。
Objective To study the effect of microfilament function on the expression of collagenase and tissue inhibitor of metalloproteinase - 1 mRNA in normal skin and hypertrophic scar fibroblasts. Methods Cell culture, Northern blot analysis and other methods to α 32P dCPT labeled collagenase and TIMP 1 cDNA as a probe, detection of normal skin and hypertrophic scar fibroblasts collagenase and metalloproteinase tissue inhibitor 1 mRNA expression level changes. Results Cytochalasin B destroyed the microfilaments, the normal skin and hypertrophic scar fibroblasts collagenase and TIMP 1 mRNA levels were significantly increased. Conclusion The actin cytoskeleton may be involved in the regulation of the extracellular matrix synthesis of fibroblasts at the gene transcription level.
