采用放射配基结合法检定了大鼠心肌细胞膜α_1及β受体,研究了离体大鼠心肌α_1及β受体在缺血再灌注损伤时的变化及预处置、ACEI的影响。结果表明,2种受体的最大结合容量在缺血40分钟、再灌注4分钟后明显升高。β受体数目的这种升高趋势可持续到再灌注20分钟,而同样条件下α_1受体数目呈恢复趋势;经预处置及ACEI保护组的2种受体的Bmax均明显低于未处置组及非ACEI保护组。各组的受体亲和力无显著变化。上述结果证明预处置、ACEI对缺血/再灌注损伤心肌有明显的保护作用。
Radioimmunoassay was used to determine the expression of α 1 and β receptors in rat myocardial cell membrane. The changes of α 1 and β receptors in isolated rat myocardium during ischemia / reperfusion injury and the effects of preconditioning and ACEI on them were studied. The results showed that the maximum binding capacity of the two receptors was significantly increased at 40 minutes after ischemia and 4 minutes after reperfusion. The increasing trend of β receptor number could last up to 20 minutes after reperfusion, while the number of α 1 receptors showed a recovery tendency under the same conditions. The Bmax of both receptors in preconditioning and ACEI protection groups were significantly lower than those in untreated Group and non-ACEI protection group. There was no significant difference in receptor affinity between groups. These results show that preconditioning, ACEI has a significant protective effect on myocardial ischemia / reperfusion injury.
