【目的】研究灰霉病拮抗菌K1的发酵条件。【方法】以OD600值为指标采用响应面法对其发酵培养基、发酵条件进行优化。通过Plackett-Burman实验确定酵母膏、p H、接种量为其最显著的3个因素。经过BoxBehnken实验和响应面实验得到最优发酵条件。【结果】最优发酵培养基为:淀粉1%,酵母膏1.8%,硫酸钾1.5%。最优发酵条件:p H6.8,接种量为2.8%,温度25℃,转速130 r/min,装液量100 m L/250 m L。【结论】K1发酵16 h后的OD600值可达到1.490,远高于优化前0.864,明显提高了K1的生长速率。优化后发酵液抑菌直径为49mm,高于优化前的43 mm。
【Objective】 The objective of the research was to study the fermentation conditions of Botrytis cinerea K1. 【Method】 Response surface methodology (OD600) was used as an index to optimize its fermentation medium and fermentation conditions. Yeast paste, p H, was determined by the Plackett-Burman experiment and the inoculum volume was the most significant of the three factors. After BoxBehnken experiments and response surface experiments to obtain the optimal fermentation conditions. 【Result】 The optimal fermentation medium was 1% starch, 1.8% yeast extract and 1.5% potassium sulfate. Optimal fermentation conditions: p H6.8, inoculum size 2.8%, temperature 25 ℃, rotation speed 130 r / min, liquid volume 100 m L / 250 m L. 【Conclusion】 The OD600 value of K1 after fermentation for 16 h reached 1.490, much higher than that of 0.864 before optimization, which obviously increased the growth rate of K1. After optimization, the bacteriostasis diameter of the fermentation broth was 49mm, which was higher than the 43mm before optimization.
