目的筛选出消毒实验中评价消毒因子对病毒灭活效果的指示噬菌体。方法比较研究噬菌体T4、φχ174D和f2对戊二醛消毒剂的抵抗力。消毒剂对噬菌体的灭活效果采用悬液定量灭活试验、中和剂鉴定试验、噬菌体的检测采用双层琼脂法。结果13000mg/L的戊二醛对噬菌体T4作用20min,或6000mg/L的戊二醛对噬菌体T4作用5min即可达到消毒水平(对噬菌体T4的灭活对数值(LIV)或噬菌体T4的对数减少值〔LRV)(log10No-log10Nt)≥4.00log10〕;22500mg/L的戊二醛对噬菌体φχ174D作用20min,或5000mg/L的戊二醛对噬菌体φχ174D作用5min达到消毒水平;34000mg/L的戊二醛对噬菌体f2作用40min,或8000mg/L的戊二醛对噬菌体f2作用10min达到消毒水平。结论上述三种噬菌体对戊二醛的抵抗力由强到弱为:噬菌体f2>噬菌体T4>噬菌体φχ174D。
Objective To screen out phage displayed by phagocytes in the disinfection experiment to evaluate the inactivation effect of disinfectant on virus. Methods The bacteriophage T4, φχ174D and f2 were tested for their resistance to glutaraldehyde disinfectant. Disinfection effect on bacteriophage phagocytosis using quantitative inactivation test, neutralizer identification test, detection of phage using double agar. Results The level of disinfection (phage T4 inactivation logarithm (LIV) or phage T4 logarithm was achieved when 13000 mg / L glutaraldehyde was applied to phage T4 for 20 min or 6000 mg / L glutaraldehyde for 5 min to phage T4 Reducing value 〔LRV (log10No-log10Nt) ≥4.00log10〕; glutaraldehyde 22500mg / L for phage φχ174D for 20min, or 5000mg / L glutaraldehyde for phage φχ174D 5min reach the level of disinfection; 34000mg / L of pentane Dialdehyde for phage f2 role 40min, or 8000mg / L of glutaraldehyde on phage f2 role 10min reached the level of disinfection. Conclusion The above three kinds of bacteriophages against glutaraldehyde resistance strong to weak as: phage f2> phage T4> phage φχ174D.
