目的:探讨四磨汤对寒冷束缚应激小鼠脑肠肽NOS的影响。方法:动物随机分组,采用寒冷束缚等多种刺激方法,制备胃肠功能紊乱小鼠模型,分别给予蒸馏水、得舒特和四磨汤口服液治疗;免疫组织化学法观察小鼠脑内NOS的阳性表达,用RT-PCR法检测结肠组织NOS mRNA表达。结果:模型组小鼠NOS阳性表达减少,平均OD值与正常组比较差异有显著性统计学意义(P<0.01);四磨汤组NOS表达增加,与模型组比较有统计学差异(P<0.05)。四磨汤可调控模型小鼠结肠组织Nos mRNA,与模型组比较差异有显著性统计学意义(P<0.01)。结论:四磨汤调节脑肠肽NOS的表达可能是其治疗功能性胃肠病的作用机制之一。
Objective: To investigate the effect of Si mill decoction on the brain-gut NOS in cold restraint stress mice. Methods: The animals were randomly divided into four groups: cold restraint and other stimulation methods to prepare mice model of gastrointestinal dysfunction, respectively, given distilled water, were Shute and Siu Tang oral solution; immunohistochemistry observed in mice brain NOS Positive expression of NOS mRNA in colonic tissue was detected by RT-PCR. Results: The NOS positive expression in the model group decreased, and the average OD value had significant difference compared with the normal group (P <0.01). The NOS expression increased in the Sihuang decoction group compared with the model group (P < 0.05). Si mill decoction can regulate the expression of Nos mRNA in colonic tissue of mice, and there is significant difference between the model group and control group (P <0.01). Conclusion: Si mill decoction can regulate the expression of gut peptide NOS, which may be one of its mechanisms in the treatment of functional gastrointestinal diseases.
