目的 观察舒林酸对结肠腺癌HT 2 9细胞增殖的影响 ,探讨其作用机制。方法 采用MTT比色法观察舒林酸对HT 2 9细胞增殖的影响 ;采用流式细胞仪检测舒林酸对HT 2 9细胞周期的影响 ,同时结合DNA电泳和透射电镜观察舒林酸对HT 2 9细胞有无促凋亡作用。结果 舒林酸可抑制HT 2 9细胞增殖 ,使G0 /G1期细胞比例增高 ,S期比例降低 ,72h后 ,HT 2 9细胞凋亡分别上升至12 .5 %、15 .4 %和 2 4 .2 % (P <0 .0 5 )。上述作用均呈时间和剂量依赖性。结论 舒林酸可抑制HT 2 9细胞增殖 ,其机制涉及影响细胞周期和诱导细胞凋亡。
Objective To observe the effect of sulindac on the proliferation of colon adenocarcinoma HT 2 9 cells and to explore its mechanism. Methods MTT assay was used to observe the effect of sulindac on the proliferation of HT29 cells. Flow cytometry was used to detect the effect of sulindac on the cell cycle of HT29 cells. Combined with DNA electrophoresis and transmission electron microscopy, 2 9 cells with or without pro-apoptotic effect. Results Sulindac inhibited the proliferation of HT 2 9 cells, increased the proportion of cells in G0 / G1 phase and decreased the proportion of S phase. After 72 hours, apoptosis of HT 2 9 cells increased to 12.5%, 15.4% and 24%, respectively .2% (P <0. 05). The above effects were time-and dose-dependent. Conclusion Sulindac inhibits the proliferation of HT 2 9 cells by affecting the cell cycle and inducing apoptosis.
