利用响应面分析法对葛花与枳椇子中总黄酮的提取工艺进行优化.通过单因素试验分别考察原料比、粉碎粒度、料液比和提取时间对总黄酮得率的影响.选择适当的试验因素和水平,利用Design Expert软件和Box-Behnken设计法设计响应面试验,对各个因素的显著性以及交互作用进行分析.结果表明:葛花与枳椇子中总黄酮的提取工艺最佳条件为原料比(葛花与枳椇子)2.2∶1,不粉碎,料液比1∶21,时间85min,在该条件下验证,总黄酮得率可达到8.31mg/g,接近预测值8.37mg/g.最优提取条件下的提取次数试验表明,提取2次效果最好.
Response surface methodology was used to optimize the extraction of total flavonoids from Pueraria lobata and Hovenia dulcis Thunb. The effects of ratio of raw materials, comminution size, solid-liquid ratio and extraction time on the yield of total flavonoids were investigated by single factor experiments, respectively. The factors and levels of test were used to design the response surface test with Design Expert software and Box-Behnken design method to analyze the significance of each factor and the interaction.The results showed that the optimum conditions for the extraction of total flavonoids from Pueraria lobata and Hovenia Dulcis Thunb As raw material ratio (Gehua and Hovenia dulcis Thunb) 2.2: 1, without pulverization, solid-liquid ratio 1:21, time 85min, under the conditions to verify the total flavonoid yield can reach 8.31mg / g, close to the predicted value of 8.37mg / g. The optimal extraction conditions under the number of tests showed that the extraction of 2 times the best.
