目的观察Rho激酶特异抑制剂Y-27632对体外培养的细胞滋养细胞侵袭能力的影响。方法2006年3月至8月在中国医科大学附属第二医院收集妊娠6~8周行人工流产术的健康妇女绒毛组织,通过免疫组化法检测细胞滋养细胞中RhoA和ROCKⅡ蛋白的表达;利用原代细胞培养技术,采用MTT、Transwell体外浸润实验和细胞运动实验检测Rho激酶抑制剂Y-27632对细胞滋养细胞生长和浸润运动能力的影响。结果在细胞滋养细胞有RhoA和ROCKⅡ蛋白的表达,原代培养的细胞滋养细胞经Y-27632处理后,侵袭及运动能力明显下降。结论Rho/Rho激酶信号转导系统对细胞滋养细胞侵袭有重要作用,可能参与由胎盘植入异常引起的妊娠相关疾病。
Objective To observe the effect of Y-27632, a Rho-kinase inhibitor, on the invasive ability of cytotrophoblasts in vitro. Methods From March to August 2006, we collected chorionic villi of healthy women who underwent induced abortion from 6th to 8th week of pregnancy in the Second Affiliated Hospital of China Medical University from March 2006 to August, and detected the expression of RhoA and ROCKⅡ protein by immunohistochemistry. The effects of Rho kinase inhibitor Y-27632 on the growth and infiltration of cytotrophoblasts were detected by MTT assay, Transwell in vitro assay and cell motility assay. Results The expression of RhoA and ROCKⅡprotein in cytotrophoblast cells. The primary culture of cytotrophoblast cells treated with Y-27632 significantly reduced the invasion and exercise capacity. Conclusion The Rho / Rho kinase signaling system plays an important role in cytotrophoblast invasion and may be involved in pregnancy-related diseases caused by abnormal placenta accreta.
