目的探讨来曲唑对大鼠子宫内膜异位症(EM)模型异位病灶中血管内皮生长因子(VEGF)及微血管密度(MVD)表达的影响,及其治疗EM的作用机制。方法行自体移植建立EM大鼠模型,将建模成功大鼠随机分为来曲唑组及盐水对照组,比较口服来曲唑或盐水前后EM大鼠异位病灶体积改变,并采用免疫组化法检测各组异位内膜VEGF及MVD的表达变化。结果来曲唑组异位病灶体积明显缩小或萎缩,与对照组比较差异有统计学意义(P<0.05)。来曲唑治疗后VEGF、MVD在异位内膜中表达降低(P均<0.05)。结论来曲唑对大鼠异位囊肿的体积影响明显,短时间内就能起到明显效果。可能通过降调VEGF、MVD在异位内膜组织中的表达而发挥治疗作用。
Objective To investigate the effect of letrozole on the expression of vascular endothelial growth factor (VEGF) and microvessel density (MVD) in ectopic endometriosis of rats with endometriosis (EM) and its mechanism of action in the treatment of EM. Methods EM rats were established by autograft. The rats were randomly divided into letrozole group and saline control group. The volume of ectopic lesion in EM rats before and after oral letrozole or saline were compared. Immunohistochemistry The changes of VEGF and MVD in ectopic endometrium in each group were detected by immunohistochemistry. Results Letrozole group ectopic lesion volume was significantly reduced or atrophy, compared with the control group, the difference was statistically significant (P <0.05). The expression of VEGF and MVD in ectopic endometrium decreased after letrozole treatment (all P <0.05). Conclusion Letrozole has significant effect on the volume of ectopic cysts in rats and can play a significant role in short time. It may play a therapeutic role by downregulating the expression of VEGF and MVD in ectopic endometrial tissues.
