目的观察米非司酮对卵巢癌细胞系SKOV3增殖和癌胚抗原相关黏附分子6(CEACAM6)表达的影响。方法不同浓度米非司酮(空白对照、1×10-7、1×10-6、1×10-5、5×10-5mol/L)干预卵巢癌细胞系SKOV324、48、72h;四甲基偶氮唑蓝(MTT)法检测米非司酮对SKOV3的抑制作用;逆转录聚合酶链反应(RT-PCR)和蛋白印迹(Westernblot)检测不同浓度米非司酮干预下CEACAM6mRNA和蛋白表达水平的差异,并进行半定量分析。结果与空白对照组比较,1×10-7mol/L和1×10-6mol/L米非司酮对SKOV3的生长没有明显的抑制作用(P>0.05);5×10-5mol/L米非司酮显著抑制SKOV3的生长(P<0.01),并降调SKOV3细胞中CEACAM6mR-NA和蛋白的表达水平(P<0.05)。结论米非司酮可明显抑制SKOV3的体外增殖,其作用机制可能与抑制CEACAM6的表达有关。
Objective To observe the effect of mifepristone on the proliferation of ovarian cancer cell line SKOV3 and the expression of carcinoembryonic antigen-6 (CEACAM6). Methods Mifepristone (blank control, 1 × 10-7, 1 × 10-6, 1 × 10-5 and 5 × 10-5mol / L) was used to treat ovarian cancer cell line SKOV324,48,72h. The inhibitory effect of mifepristone on SKOV3 was detected by MTT assay. The expression of CEACAM6 mRNA and protein under different concentrations of mifepristone was detected by reverse transcription polymerase chain reaction (RT-PCR) and Western blotting Horizontal differences, and semi-quantitative analysis. Results Compared with the blank control group, 1 × 10-7mol / L and 1 × 10-6mol / L mifepristone had no significant inhibitory effect on the growth of SKOV3 (P> 0.05); 5 × 10-5mol / L Cholesterol significantly inhibited the growth of SKOV3 (P <0.01), and decreased the expression of CEACAM6mR-NA and protein in SKOV3 cells (P <0.05). Conclusion Mifepristone can significantly inhibit the proliferation of SKOV3 in vitro and its mechanism may be related to the inhibition of CEACAM6 expression.
