目的:探讨前列安治疗前列腺增生症的作用机制。方法:采用大鼠去势后注射丙酸睾酮致前列腺增生法造模。分5组,即正常组、模型组、前列安低剂量0.1 g.kg-1组、前列安高剂量0.2 g.kg-1组、癃闭舒0.2 g.kg-1组,ig给药30 d后处死。摘取大鼠前列腺并用电光分析天平称重,获取前列腺指数,同时制作光镜和免疫组化标本,对每张免疫组化片进行显微定性分析并观察光镜下前列腺细胞形态变化。结果:3个治疗组大鼠前列腺重、前列腺指数及碱性成纤维生长因子(bFGF)表达水平均明显低于模型组,其中尤以前列安高剂量组最为明显。模型组大鼠前列腺组织见明显的病理性改变,前列安低剂量组大鼠前列腺组织的病理性改变较模型组减轻,高剂量组和癃闭舒组大鼠前列腺组织的病理改善最为明显。结论:前列安可降低大鼠前列腺湿重,修复增生的病理损伤,其作用机制可能与前列安抑制前列腺组织中bFGF释放有关。
Objective: To investigate the mechanism of Qianliean in the treatment of benign prostatic hyperplasia. Methods: Rats were castrated and injected with testosterone propionate to make the model of benign prostatic hyperplasia. The rats were randomly divided into 5 groups: normal group, model group, Qianliean low dose 0.1 g.kg-1 group, Qianliean high dose 0.2 g.kg-1 group, Shugan Shu Shu 0.2 g.kg-1 group, ig administration 30 d after sacrifice. Prostate of rats were taken out and weighed by electro-optical balance to obtain Prostate index. At the same time, light microscope and immunohistochemistry specimens were made. Immunohistochemical analysis was performed on each immunohistochemical piece and the morphological changes of prostatic cells under light microscope were observed. Results: The expression of prostate weight, prostate index and basic fibroblast growth factor (bFGF) in the three treatment groups were significantly lower than those in the model group, especially in the Qianliean high dose group. The pathological changes of prostate tissue were observed in the model group. The pathological changes of the prostate tissue of the Qianliean low dose group were alleviated compared with the model group, and the pathological improvement of the prostate tissue was the most obvious in the high dose group and the Shuo Shutushu group. Conclusion: Qianlie An can reduce the wet weight of the prostate, repair the pathological damage of hyperplasia, the mechanism may be related to the prostaglandin suppression prostate bFGF release.
