本研究应用3H-Tdr经生物合成法制备玉米3H-DNA,借助微观放射自显影(RAG)技术,按常规方法进行操作,探讨玉米DNA经花粉管通道进入水稻胚囊的可行性。结果表明,生物合成标记的DNA放射性活度满足RAG要求,花柱切口部位存在3H-DNA,在部分花柱切片中发现自显影银粒,但胚囊内未能检验到。
In this study, 3H-Tdr biosynthesis method was used to prepare maize 3H-DNA. The feasibility of using maize DNA to enter the rice blastocyst through the pollen tube pathway was studied by means of microscopic autoradiography (RAG). The results showed that the radioactivity of the biosynthetic marker DNA meets the RAG requirements. 3H-DNA was present at the cuttings, and self-developed silver particles were found in some of the style segments. However, the embryo sac failed to be detected.
