目的 :观察肝纤维化形成过程中 α平滑肌肌动蛋白 (α- SMA)与门脉压力变化及两者的关系。方法 :采用二甲基亚硝胺 (DMN)诱发大鼠肝纤维化模型 ,共 4周。分别于造模开始后 2 d,1、2、3、4、5、6、8、12及 2 4周末观察大鼠肝纤维化程度、α- SMA表达与门脉压力。结果 :造模过程中肝纤维化程度逐渐加重 ,至第 4周末最为明显 ,其后逐渐减轻 ;α- SMA表达与肝纤维化程度的变化相一致 ,两者呈正相关 ;门脉压力的变化与纤维化程度及 α- SMA表达均呈正相关。结论 :肝星状细胞 (HSC)的活化在肝纤维化形成中起着重要作用 ,并对门脉压力起调节作用。
Objective: To observe the changes of α-SMA and portal pressure in the process of hepatic fibrosis and their relationship. Methods: The rat model of hepatic fibrosis was induced by dimethylnitrosamine (DMN) for 4 weeks. The degree of hepatic fibrosis, α-SMA expression and portal pressure were observed at 2 days, 1, 2, 3, 4, 5, 6, 8, 12 and 24 weeks after the onset of modeling. Results: The degree of hepatic fibrosis gradually increased during the modeling process, and it became the most obvious at the end of the 4th week, and then gradually decreased. The expression of α-SMA was consistent with the changes of liver fibrosis, and the changes of portal pressure and The degree of fibrosis and the expression of α-SMA were positively correlated. Conclusion: The activation of hepatic stellate cells (HSC) plays an important role in the formation of hepatic fibrosis and regulates portal pressure.
