目的探讨半胱氨酸蛋白酶抑制剂C(CystatinC)在胃肠道肿瘤组织中的表达及其在肿瘤发生发展过程中的意义。方法运用免疫组化及WesternBlotting的方法分别对肿瘤组织及其癌旁组织中CystatinC的表达进行分析,并运用聚合酶链反应测定(ELISA)方法检测肿瘤组织及其癌旁组织中CystatinC的含量。结果(1)免疫组化结果显示CystatinC主要表达在肿瘤细胞的胞浆内,正常腺体中有微弱表达,且胃肠道肿瘤组织中的阳性百分比(75%)显著高于癌旁组织的阳性百分比(46·88%)(χ2=6·825,P<0·01);(2)WesternBlotting的结果证实CystatinC在肿瘤组织中的表达明显高于癌旁组织;(3)ELISA定量测定肿瘤组织中CystatinC的含量高于癌旁组织,[符号秩和检验(WilcoxonSignedRanksTest),S=33·00,P<0·05];(4)CystatinC在高分化肿瘤中的含量高于低分化肿瘤(Mann-WhitneyTest,S=13·00,P<0·05)。结论CystatinC在胃肠道肿瘤组织中表达增高并与肿瘤的恶性程度存在相关性,提示CystatinC在胃肠道肿瘤的发生发展过程中起重要作用。
Objective To investigate the expression of cystatin C in gastrointestinal tumor tissue and its significance in the development of tumor. Methods The expression of Cystatin C in tumor tissue and adjacent tissues was analyzed by immunohistochemistry and Western Blotting. Cystatin C in tumor tissue and adjacent tissues was detected by polymerase chain reaction (ELISA). Results (1) Immunohistochemical results showed that CystatinC was mainly expressed in the cytoplasm of tumor cells with weak expression in normal glands, and the positive percentage (75%) in gastrointestinal tumor tissues was significantly higher than that in paracancerous tissues (46.88%) (χ2 = 6 · 825, P <0.01). (2) The results of Western Blotting confirmed that the expression of CystatinC in tumor tissues was significantly higher than that in paracancerous tissues. (3) CystatinC was higher than that in paracancerous tissues (Wilcoxon signed Ranks test, S = 33.00, P <0.05). (4) Cystatin C levels in well-differentiated tumors were higher than those in poorly differentiated tumors - Whitney Test, S = 13.00, P <0.05). Conclusions CystatinC is highly expressed in the gastrointestinal tract and correlates with the malignancy of the tumor, suggesting that Cystatin C plays an important role in the development of gastrointestinal tumors.
