目的:制备川牛膝化学拆分组分,并研究川牛膝水煎液及其各化学拆分组分对免疫调节的影响。方法:采用水煎煮、醇沉、大孔吸附树脂柱色谱分离技术的联合应用,将川牛膝水煎液的化学成分拆分为成分之间互不交叉的以下几部分:多糖组分,20%乙醇洗脱组分,甾酮组分和脂肪油组分,对其进行化学特征研究并运用HPLC对各化学拆分组分进行互不交叉性研究。考察川牛膝水煎液及其各化学拆分组分对小鼠碳粒廓清指数和免疫器官的影响及对小鼠血清溶血素抗体生成的影响。结果:建立的川牛膝化学成分拆分方法可行、稳定且重复性好,能够实现全成分拆分,各拆分组分间化学成分基本无交叉;川牛膝具有一定的免疫增强作用。结论:发挥免疫增强作用的药效物质基础为多糖组分和甾酮组分。
OBJECTIVE: To prepare chemical splitting components of Achyranthes bidentata and to study the effects of Achyranthes bidentata decoction and its chemical constituents on immune regulation. Methods: The combined application of water decoction, alcohol precipitation, macroporous resin column chromatography separation technology, the chemical composition of the decoction of Radix Achyranthis Bidentata split into the following components do not cross each other: polysaccharide components, 20% ethanol elution fraction, steroid fraction and fatty oil fraction were prepared and their chemical characteristics were studied. The chemical composition of each fraction was studied by HPLC. The effects of decoction of Radix Achysthesiae and its chemical constituents on carbon clearance index and immune organs in mice and the effect on serum hemolysin antibody production in mice were investigated. Results: The established chemical constituents of Achyranthes bidentata were feasible, stable and reproducible, and could achieve the resolution of the whole components. The chemical components of the separated components were almost non-crossover. Achyranthes bidentata had some immune enhancement effects. CONCLUSION: The pharmacological substance that exerts immune enhancement is polysaccharide component and steroid component.
