目的:探索抑制着丝粒蛋白F(CENP-F)表达对人胶质母细胞瘤细胞系U251增殖的影响。方法:培养U251细胞,转染抑制片段,检测空白对照组(未做任何处理,Blank组),阴性对照组(转入空白质粒组,NC组)及转入抑制片段组(si-CENPF组,根据转入的干扰片段分si-CENPF#1组、si-CENPF#2组、si-CENPF#3组)各组CENP-F m RNA的表达。挑选抑制最明显的片段进行Western-botting进一步检测CENP-F蛋白水平的表达。行MTT检测转染细胞组与空白对照组及阴性对照组的细胞增殖力。结果:干扰片段能抑制U251细胞系CENP-F m RNA水平及蛋白水平的表达,同时能抑制U251细胞的增殖。结论 :抑制CEAFP的表达能降低人胶质母细胞瘤细胞系U251的增殖。
Objective: To explore the effect of CENP-F inhibition on proliferation of human glioblastoma cell line U251. METHODS: U251 cells were cultured and transfected with inhibitory fragments. The blank control group (Blank group), negative control group (transfected into blank plasmid group, NC group) and the transfected inhibition group (si-CENPF group, The expression of CENP-F m RNA in each group was divided into groups according to the interfering fragments transfected by si-CENPF # 1 group, si-CENPF # 2 group and si-CENPF # 3 group. The most significant fragment was selected for Western-botting for further detection of CENP-F protein expression. MTT test transfected cells and blank control group and negative control group cell proliferation. Results: The interference fragment could inhibit the expression of CENP-F m RNA and protein in U251 cell line and inhibit the proliferation of U251 cell line. Conclusion: Inhibition of CEAFP expression can reduce the proliferation of human glioblastoma cell line U251.
