目的 探讨玻璃体内插入外周神经和玻璃体内注射CPT 环磷酸腺苷对轴突损伤的视网膜节细胞再生的影响。 方法 2 0只成年金黄地鼠随机分 4组 ,每组 5只。切断视神经 (ON)并缝接坐骨神经为再生对照组(attachedgraft,AG) ;ON切断并缝接坐骨神经后再于玻璃体内注射CPT cAMP和 或插入一小段自体坐骨神经 (SN)为实验组。采用逆行示踪标记术 ,观察视网膜平铺片节细胞数。 结果 1 对照组 (AG) ,视网膜再生的节细胞数为 :142 8± 2 84个 ;2 外周神经组 (AG +SN) :每个视网膜再生的节细胞平均数为 2 2 2 0± 415个 ;3 CPT cAMP组 (AG+cAMP) :每个视网膜再生的节细胞平均数为 2 175± 36 4个 ;4 外周神经和CPT cAMP联合组 (AG +cAMP +SN) :每个视网膜再生的节细胞平均数为 42 5 5± 82 0个 ;其中 ,AG +SN、AG +CPT组同AG组相比存在差异显著性 (P <0 0 1) ;AG +CPT +SN组同AG、AG +CPT和AG +SN组相比均有差异显著性 (P <0 0 1)。 结论 研究结果提示玻璃体内注射CPT +cAMP或联合应用外周神经和CPT cAMP可大幅提高受损视网膜节细胞的再生。
Objective To investigate the effect of intravitreal injection of CPN and intravitreal CPT on the regeneration of retinal ganglion cells injured by axon. Methods Twenty adult golden hamsters were randomly divided into 4 groups with 5 in each. The optic nerve (ON) was cut off and the sciatic nerve was sewn to the regenerative control group (AG). ON was cut off and sutured to the sciatic nerve, then intravitreal injection of CPT cAMP and / or insertion of a short piece of autologous sciatic nerve (SN) were used as the experimental group. Retrograde tracing technique was used to observe the number of retinal laminar ganglion cells. Results 1 In control group (AG), the number of ganglion cells regenerated in retina was 142 8 ± 2 84. 2 Peripheral nerves group (AG + SN): The average number of ganglion cells regenerated per retina was 2220 ± 415 ; 3 CPT cAMP group (AG + cAMP): The average number of ganglion cells regenerated per retina was 2 175 ± 36 4; 4 Combination of peripheral nerves and CPT cAMP group (AG + cAMP + SN) The average number of cells in the AG + CPT + SN group was 42 5 5 ± 82 0, AG + SN, AG + CPT group was significantly different from AG group (P <0.01) There was significant difference between CPT group and AG + SN group (P <0.01). Conclusions The results suggest that intravitreal injection of CPT + cAMP or combination of peripheral nerve and CPT cAMP can significantly improve the regeneration of injured retinal ganglion cells.
