苜蓿根瘤菌1021中多个腺苷酸环化酶基因的存在,暗示着cAMP的重要性.本研究用全基因组DNA微阵列分析了外源cAMP对苜蓿根瘤菌的功能.分析结果表明,glnⅡ和glnK的转录水平在外源cAMP存在时有明显上调.通过测定glnⅡ和glnK启动子与lacZYA报告基因融合系统在苜蓿根瘤菌体内的活性,再次证实外源cAMP对glnⅡ和glnK启动子表达有激活作用.
The presence of multiple adenylate cyclase genes in S. meliloti 1021 implies the importance of cAMP.In this study, the function of exogenous cAMP on alfalfa rhizobia was analyzed using genome-wide DNA microarrays.The results showed that gln Ⅱ and The transcriptional level of glnK was significantly upregulated in the presence of exogenous cAMP, and the activation of glnⅡ and glnK promoters was confirmed by exogenous cAMP by measuring the activity of gln Ⅱ and glnK promoter and lacZYA reporter gene fusion system in Rhizobium meliloti.
