目的:初步研究补肾益精法中药复方对硬皮病成纤维细胞SMAD3/FLI1“平衡”的影响。方法:体外培养建立硬皮病组和正常对照组皮肤成纤维细胞系;使用补肾益精法中药复方含药血清与对照血清处理两组细胞;采用实时定量RT-PCR技术测定I型前胶原COLⅠ-α2与TGF-β途径中SMAD3、FLI1等因子的基因转录水平。结果:补肾益精法可不同程度地减少硬皮病成纤维细胞COLⅠ-α2、SMAD3表达,上调FLI1基因表达,使SMAD3/FLI1比值趋向健康对照组水平。结论:补肾益精法对硬皮病成纤维细胞表型的影响可能与TGF-β途径中SMAD3/FLI1“平衡”有关。
Objective: To study the effect of Bushen Yijing method on SMAD3 / FLI1 balance of scleroderma fibroblasts. Methods: The skin fibroblast cell lines were established by culturing scleroderma in vitro and the normal control group. The two groups of cells were treated with Bushen Yijing Pharmacotherapy serum and the control serum. Real-time quantitative RT-PCR was used to detect COLⅠ -α2 and TGF-β pathway SMAD3, FLI1 and other factors of gene transcription level. Results: Bushenyijing method can reduce the expression of COLⅠ-α2 and SMAD3 in scleroderma fibroblasts to some extent, and up-regulate the expression of FLI1 gene, so that the ratio of SMAD3 / FLI1 tends to the level of healthy control group. Conclusion: The effect of Bushen Yijing Method on the phenotype of fibrotic cells of scleroderma may be related to the SMAD3 / FLI1 balance in TGF-β pathway.
