目的:探讨脱氢表雄酮(DHEA)对实验性自身免疫性神经炎(EAN)大鼠细胞免疫反应的影响。方法:21只Lewis大鼠随机分为DHEA 0.5mg治疗组、2mg治疗组和对照组,治疗组于注射牛周围神经髓磷脂(BPM)抗原乳剂后第5天开始每日皮下注射DHEA,对照组皮下注射相同体积的DHEA溶媒。疾病高峰期检查坐骨神经IFN-γ、TNF-α阳性反应细胞,检测引流淋巴结和脾脏单个核细胞增殖反应,检测培养上清TNF-α、IFN-γ、IL-10含量。结果:两种剂量DHEA治疗组均能减少坐骨神经IFN-γ、TNF-α阳性反应细胞数目(P<0.05),抑制BPM刺激T淋巴细胞增殖(P<0.05),降低培养上清中IFN-γ、TNF-α水平(P<0.05)。各组间IL-10水平差异无统计学意义(P>0.05)。结论:DHEA可抑制EAN大鼠自身反应性T淋巴细胞增殖和促炎性细胞因子过度表达,抑制细胞免疫反应。
Objective: To investigate the effect of dehydroepiandrosterone (DHEA) on cellular immune response in experimental autoimmune neuritis (EAN) rats. Methods: Twenty-one Lewis rats were randomly divided into DHEA 0.5 mg treatment group, 2 mg treatment group and control group. The treatment group received subcutaneous injection of DHEA daily on day 5 after injection of bovine peripheral nerve myelin (BPM) Subcutaneous injection of the same volume of DHEA vehicle. The sciatic nerve IFN-γ and TNF-α positive cells were detected during the peak period of disease. The proliferation reaction of draining lymph node and spleen mononuclear cells were detected. The content of TNF-α, IFN-γ and IL-10 in the culture supernatant was detected. Results: Both doses of DHEA could reduce the number of IFN-γ and TNF-α-positive cells (P <0.05), inhibit the proliferation of T lymphocytes stimulated by BPM (P <0.05) and reduce the levels of IFN-γ , TNF-α levels (P <0.05). There was no significant difference in the level of IL-10 among the three groups (P> 0.05). Conclusion: DHEA can inhibit EAN rat autoreactive T lymphocyte proliferation and proinflammatory cytokine overexpression, inhibition of cellular immune response.
