目的 培养大鼠肠黏膜微血管内皮细胞,为内皮细胞的体外研究提供实验材料。 方法 利用机械 吹打法和酶消化法分别分离出肠绒毛固有层,采用植块培养法培养原代内皮细胞,依次以局部消化法和差速黏附 法进行纯化,细胞鉴定采用第Ⅷ因子相关抗原免疫荧光检测和硝酸银染色。 结果 成功分离培养出大鼠的肠黏 膜微血管内皮细胞,纯化后的细胞可传到18代以上。 结论 为肠黏膜微血管内皮细胞的体外培养建立了一种 操作简单、成本低廉的方法。
Objective To culture rat intestinal mucosa microvascular endothelial cells and provide experimental materials for in vitro studies of endothelial cells. Methods The lamina propria lamina of E.coli was isolated by mechanical blowing method and enzymatic digestion method. Primary cultured endothelial cells were cultured by explant culture method, and then purified by local digestion and differential adhesion method. The cells were identified by factor Ⅷ related antigen Immunofluorescence and silver nitrate staining. Results The intestinal mucosa microvascular endothelial cells of rats were successfully isolated and cultured, and the purified cells could be passaged to more than 18 passages. Conclusions A simple and inexpensive method for the culture of intestinal mucosal microvascular endothelial cells in vitro was established.
