目的:通过观察痹痛定分散片对大鼠AA模型血清TGF-β1含量及滑膜组织病理形态学的影响,从细胞因子角度以及组织病理形态学改变探讨其的作用机制。方法:60只SD大鼠,随机分为6组,以Freund s完全佐剂在每只鼠右后足跖皮内注射0.05ml致炎,复制佐剂性关节炎(AA)SD大鼠模型。造模后第0、3、6、18、24h各再次测定其右足垫厚度,以造模前后厚度之差值为肿胀程度;采血检测血清中TGF-β1含量,并取病理组织进行相应检测及积分评价。结果:各治疗组与模型组比较能明显改善足肿胀度;且能明显提高血清中TGF-β1含量,Ⅲ、Ⅳ、Ⅴ、Ⅵ组与模型组比较有较显著性差异(P<0.01),Ⅳ、Ⅴ、Ⅵ组与正常组比较血清TGF-β1含量有较显著性差异(P<0.01),各治疗组能有效减轻AA模型滑膜组织病理形态的改变。结论:痹痛定分散片能有效治疗AA炎症,明显改善AA大鼠滑膜组织病理形态学改变,改善关节局部以及整体状态,提高血清TGF-β1含量,有助于机体恢复免疫稳定状态,具有免疫调节效应。
OBJECTIVE: To observe the effect of Biantongding disperse tablet on the serum TGF-β1 content and synovial tissue pathomorphology of AA model in rats, and to explore its mechanism of action from the perspective of cytokines and histopathological changes. Methods: Sixty Sprague-Dawley rats were randomly divided into 6 groups and Freund’s complete adjuvant was used to immunize the right hind paw of each rat with 0.05 ml of inflammation. SD rat model of adjuvant arthritis (AA) was duplicated. At the 0th, 3rd, 6th, 18th, and 24th hours after modeling, the thickness of the right footpad was measured again. The difference between the thickness before and after the modeling was the degree of swelling; the blood was taken for the detection of serum TGF-β1 content, and the pathological tissue was taken for the corresponding test. Integral evaluation. Results: Compared with the model group, each treatment group can significantly improve the degree of foot swelling; and can significantly increase the serum TGF-β1 content, III, IV, V, VI group compared with the model group there is a significant difference (P <0.01). The serum levels of TGF-β1 in the IV, V and VI groups were significantly higher than those in the normal group (P<0.01). Each treatment group was able to effectively reduce the pathological changes in the synovial tissue of the AA model. Conclusion: Bitongding dispersible tablets can effectively treat AA inflammation, significantly improve the pathomorphological changes of synovial tissue of AA rats, improve the local and overall state of joints, increase serum TGF-β1 content, and help the body to restore immune stable state.
