目的探讨研究大鼠骨髓间充质干细胞的体外分离、培养及鉴定。方法利用流式细胞检测技术,检测了分离培养的贴壁细胞CD29、CD90和CD45表面抗原的表达。结果本实验通过流式细胞检测,发现CD29阳性率为91.9%;CD45阳性率为6.9%;CD90阳性率51.3%;CD29/CD45阳性率为7.4%;CD90/CD45阳性率为3.6%。结论本研究通过贴壁筛迭法和全骨髓培养法得到的细胞是骨髓中不同于造血系细胞的另一种细胞成分,应为间充质干细胞,不是造血系细胞。
Objective To study the isolation, culture and identification of rat bone marrow mesenchymal stem cells in vitro. Methods Flow cytometry was used to detect the expression of CD29, CD90 and CD45 surface antigen on cultured adherent cells. Results The results of flow cytometry showed that the positive rate of CD29 was 91.9%, the positive rate of CD45 was 6.9%, the positive rate of CD90 was 51.3%, the positive rate of CD29 / CD45 was 7.4% and the positive rate of CD90 / CD45 was 3.6%. CONCLUSIONS: The cells obtained by adherence screening and whole bone marrow culture in this study are bone marrow cells that are different from hematopoietic cells. They should be mesenchymal stem cells, not hematopoietic cells.
