目的研究预感染旋毛形线虫1周后C57BL/6小鼠抵御致死型约氏疟原虫攻击感染的能力。方法实验组在小鼠感染旋毛形线虫1周后感染约氏疟原虫,对照组单纯感染约氏疟原虫,观察小鼠原虫血症及生存率的变化。采用流式细胞仪技术检测脾细胞悬液中T细胞数量,酶联免疫吸附试验定量检测脾细胞培养上清中细胞因子,Real-Time PCR法检测转录因子的表达情况。结果与对照组相比,实验组小鼠原虫血症水平降低,差异有统计学意义(P<0.05),生存率并未发生改变。实验组小鼠于感染后3 d,CD4+IFN-γ+T细胞及其效应分子IFN-γ,转录因子T-bet相对表达量明显升高,与对照组比较差异有统计学意义(P<0.05)。结论旋毛形线虫预感染1周可增强C57BL/6小鼠对致死型约氏疟原虫的早期抗感染能力。
Objective To investigate the ability of C57BL / 6 mice to resist lethal challenge with Plasmodium yoelii against infection 1 week after pre-infection with Trichina. Methods The experimental group infected with Plasmodium yoelii after 1 week in mice infected with Trichinella spiralis and the control group was infected with Plasmodium yoelii only to observe the changes of parasitemia and survival rate in mice. The number of T cells in the spleen cell suspension was detected by flow cytometry. The cytokines in the spleen cell culture supernatants were detected by enzyme-linked immunosorbent assay (ELISA), and the expression of transcription factors were detected by Real-Time PCR. Results Compared with the control group, the level of parasitemia in the experimental group was decreased, the difference was statistically significant (P <0.05), and the survival rate did not change. At 3 d after infection, the relative expression of IFN-γ and T-bet of CD4 + IFN-γ + T cells and their effector molecules in experimental mice increased significantly compared with the control group (P < 0.05). Conclusion Pretreatment with Trichomonal nematodes for 1 week can enhance the early anti-infective ability of lethal Plasmodium yoelii in C57BL / 6 mice.
