目的观察大鼠卡氏肺孢子菌肺炎(PCP)模型建立过程中鼠肺组织学、病原学、外周血肿瘤坏死因子-α(TNF-α)、可溶性粘附分子-1(sICAM-1)和白介素-8(IL-8)含量的动态变化。方法建立大鼠PCP模型,观察建模2、4、6、8和10周时大鼠的发病状况、体重、肺重改变、鼠肺组织学变化、鼠肺印片中卡氏肺孢子菌(PC)包囊形态和增殖数量,并检测大鼠外周血血清中TNF-α、sICAM-1和IL-8的含量。结果建模2周时就可在大鼠肺脏印片中发现PC包囊,但建模6周和8周时PC包囊增殖无显著差异(P>0.05)。建模4周时大鼠出现明显的PCP病征和鼠肺组织学变化,大鼠外周血血清中TNF-α、sICAM-1和IL-8的含量也在建模4周时出现升高,其中TNF-α和sICAM-1升高迅猛,IL-8升高缓慢。结论大鼠建模4周后可出现较为明显的PCP相关病征,而建模6周和8周之间可能存在肺孢子菌与宿主相互作用的相持时期。
Objective To observe the changes of tumor necrosis factor-α (TNF-α), soluble adhesion molecule-1 (sICAM-1) and tumor necrosis factor-α (TNF-α) in rat lung during the establishment of rat model of Pneumocystis carinii pneumonia Dynamic changes of interleukin-8 (IL-8) content. Methods The rat model of PCP was established. Morphological changes, body weight, lung weight and histopathological changes of rat lung were observed at 2, 4, 6, 8 and 10 weeks after modeling. Pneumocystis carinii PC), and the levels of TNF-α, sICAM-1 and IL-8 in peripheral blood of rats were detected. Results PC cysts were found in rat lung impressions at 2 weeks after modeling. However, there was no significant difference in PC cytoplasm proliferation between 6 and 8 weeks after modeling (P> 0.05). At 4 weeks after injection, significant PCP symptoms and histological changes of rat lung were observed in rats. The contents of TNF-α, sICAM-1 and IL-8 in peripheral blood of rats also increased at 4 weeks after modeling TNF-α and sICAM-1 increased rapidly, IL-8 increased slowly. Conclusion There is obvious PCP related symptoms after 4 weeks of modeling in rats, and there may be stalemate of interaction between Pneumocystis and host during 6 weeks and 8 weeks after modeling.
